Use and maintenance of refrigerated centrifuge

The cryogenic separation technology of cryogenic refrigerated centrifuge is an indispensable means in molecular biology research. The separation of gene fragments, the precipitation and recovery of enzyme proteins, and the separation and preparation of other biological samples are all inseparable from cryogenic centrifugation. Therefore, cryogenic refrigerated centrifuges have become an essential tool in molecular biology research.

Instructions:

1. The centrifuge should be placed on a level solid floor or platform and strive to keep the machine in a horizontal position to avoid machine vibration during centrifugation.

2. Turn on the power switch, install the required rotor as required, and place the sample with the balance of the tray balance in advance on the rotor sample holder (the centrifuge tube must be balanced with the sample) and close the cover.

3. Press the function selection button to set the requirements: temperature, speed, time, acceleration and deceleration. The machine with computer control also needs to press the store button to remember the input information.

4. Press the start button, the refrigerated centrifuge will perform the above parameters to operate, and automatically shut down to the scheduled time.

5. After the centrifuge has completely stopped rotating, open the cover, take out the centrifuge sample, and wipe the rotor and the inner wall of the machine cavity with a soft and clean cloth. After the temperature in the centrifuge chamber is balanced with the room temperature, the cover can be closed.

Precautions:

1. The body should always be in a horizontal position, the voltage of the external power supply system should be matched, and a good grounding wire is required.

2. Before starting the machine, check whether the rotor is installed securely, and whether there is any foreign matter falling into the machine cavity.

3. The sample should be pre-equilibrated. When using a centrifuge tube , the centrifuge tube and the sample should be balanced at the same time.

4. When the volatile or corrosive liquid is centrifuged, use a centrifuge tube with a cover to ensure that the liquid does not leak, so as not to corrode the machine cavity or cause an accident.

5. Wipe the frozen centrifuge chamber with light action to avoid damage to the temperature sensor inside the chamber.

6. Record the usage status after each operation, and regularly check the performance of the machine.

7. If abnormal phenomena are found during the centrifugation process, the power should be turned off immediately and reported to the relevant technical personnel for inspection.

The speed of the centrifuge is generally expressed in terms of how many revolutions per minute in the previous experimental data. Since the centrifugal force is not only a function of the rotational speed but also a function of the centrifugal radius, that is, the longer the centrifugal radius, the greater the centrifugal force generated. Therefore, it is not scientific enough to express centrifugal force only at the rotational speed. In recent years, it has been reasonable to use relative centrifugal force (RCF). In the international data, it has been changed to use relative centrifugal force .
