Fixed cell processing:
1. Collect the cell suspension, and the number of cells is 2 × 10^5 ~ 1 × 10^6. Centrifuge the cells by centrifugation, remove the supernatant, and tap the tube.
Resuspend the pellet with the remaining liquid; add 1 ml of PBS at room temperature;
2. All the cells were slowly dropped into 3 ml of ice ethanol (-20 ° C), and vortexed at high speed while dropping.
The cells can be stored for several months at -20 °C.
3. Centrifuge the cells by centrifugation, discard the ethanol, tap the tube to loosen the pellet, add 2~5ml of PBS at room temperature, and let stand for 15min to rehydrate the cells.
Discard the supernatant by centrifugation.
4. Add 1 mL of Reagent A and vortex for 5 to 10 s. Incubate for 30 min at room temperature.
Live cell processing:
1. Collect the cell suspension, and the number of cells is 2 × 10^5 ~ 1 × 10^6. The cells were pelleted by centrifugation and the supernatant was removed.
Wash once with PBS and discard the supernatant.
2. Add 1 mL of Reagent A and 10 μL of Reagent B and vortex for 5 to 10 s. Incubate for 30 min at room temperature.
Flow analysis: The incubated cell suspension was applied directly to the machine.
Fluorescence microscopy:
Centrifuge the incubated cell suspension to remove the supernatant in fresh buffer (eg PBS)
Resuspend the cells in medium.
Add the cells to the slides, cover the coverslips, and observe with a suitable filter.
