Introduction
A major challenge in integrated lipid analysis (lipidomics) is the separation of multiple lipids in biological samples. Using a single-dimensional chromatography (e.g. reverse phase or normal phase separation) of such separation can not be completed 1-5. Normal phase UPLC is based on lipid-based polar head groups for separation, while reversed-phase UPLC separates lipids 1-6 based on acyl chain length and number of double bonds. Hydrophilic Interaction Chromatography (HILIC) separation is an alternative to normal phase separation, providing better MS compatibility and less solvent toxicity 4-6 . Recently, researchers have proposed a two-dimensional separation method using HILIC and reverse UPLC to maximize liposome separation before MS detection. 5,6 Except chromatography, ion mobility method can also be 7,8 lipid plasma vapor separation according to the size and molecular shape.
In this study, the Waters® omics research platform combined with TransOmics informatics software and the HILIC-UPLC separation using ion mobility Tof MS (SYNAPT G2-S HDMS) will enable multidimensional separation of complex biological mixtures. Improve the information obtained in lipid analysis. HDMS control software and TransOmics informatics software can help achieve comparisons between biological samples.
