Analysis of migration motion of breast cancer using laser holographic cell imaging and analysis system

In recent years, the incidence of breast cancer in China has continued to increase. In some developed cities, breast cancer has ranked first among female malignant tumors. According to the type of ER of estrogen receptor, it is hormone-independent and dependent. Sexuality, research on ER has important implications for the pathogenesis, clinical treatment and prognosis of breast cancer.

ER is an important biomarker for breast cancer. During the development and growth of the mammary gland, estrogen and progesterone are regulated. ER is an estrogen receptor in breast cells. When cancer occurs, it can be used for patients with positive ER. 70-80% of the effects are achieved by endocrine therapy, and less than 10% for patients with negative ER, indicating that patients with negative ER are completely out of hormone regulation. In this study, we mainly investigated the relationship between ERα and lymphatic metastasis in breast cancer by invading lymphocytes from negative ER breast cancer cells.

Experimental Materials:

Using MCF-7 breast cancer cell line as experimental material, Cas9-ERα treated with gene editing technology, Cas9-ERα+ERα treated with matrix metalloproteinase inhibitor MG6001, and Cas9-vinculin were performed using Swedish HoloMonitor M4. The movement and migration of the cells were photographed and observed for 4 hours, once every 2 minutes. The cell migration movements of different experimental groups were compared.

The experimental results showed that the cell migration of Cas9-ERα was significantly increased compared with the control group, and five different forms of cell deformation were captured, respectively (1-5movie in the supplement). Moreover, in the experimental group of Cas9-ERa+MG6001, the metalloproteinase inhibitor did not inhibit the high invasiveness of cells after lack of ERa. The percentage of cells in five different forms was analyzed. The cell migration and cell motility rate of Cas9-vinculin was also significantly higher than that of the control group, and five different forms in the process of cell deformation were also observed (6-10movie in the supplement). The comprehensive data show that in breast cancer cells, the loss of ERα leads to a decrease in the expression of Vinculin, which directly leads to an increase in intracellular myosin light chain p-MLC, resulting in increased cell invasiveness. .
