When a gene mutation in a cell causes abnormal growth of the cell, cancer is produced. Some cancer drugs attack tumor cells by targeting mutant proteins encoded by mutant genes. However, only a few mutations that have made outstanding contributions to cancer have been identified.
Thomas Peterson of the University of Maryland and colleagues developed a new approach to data analysis. This method uses genetic data from cancer patients to identify mutations that cause cancer. Unlike previous concerns about individual gene mutations, the new approach focuses on similar mutations common to related protein families.
In particular, the new approach focuses on protein substructure mutations known as protein domains. Although these proteins are encoded by different genes, different proteins have a common protein domain. The new approach leverages existing knowledge and functions of protein domains to determine the location of protein domains that are more likely to be found in tumors.
Using this new approach, the researchers identified thousands of rare tumor mutations that occurred in the same location as other tumor protein mutations. This suggests that they may be related to cancer.
The study's senior author, Maricel Kann, said: "Perhaps only two patients have mutations in specific proteins. But when you find that it is in exactly the same position as other cancer patients' protein mutations, you will realize that these two studies The importance of a mutation."
